Detector Introduction
The UM-5000 evaporative light scattering detector is designed for use in high-performance liquid chromatography systems to analyze any compound with lower volatility than the mobile phase. The application scope of ELSD includes carbohydrates, drugs, lipids, undifferentiated fatty acids and amino acids, polymers, surfactants, nutritional supplements, and combination molecular libraries. The evaporative light scattering detector eliminates common issues found in other HPLC detectors. Differential detection is complicated by the interference of solvent front peaks, and due to its extreme sensitivity to temperature, the baseline is very unstable and incompatible with gradient elution. In addition, the response of differential detectors is not as sensitive as ELSD. Low wavelength ultraviolet detectors are plagued by baseline drift under rapidly changing gradient conditions and require the analyzed compound to have chromophores. ELSD is not subject to these restrictions. Unlike these detectors, ELSD can obtain stable baselines in the presence of multiple solvent gradients, resulting in better resolution and faster separation speed. In addition, because the response of ELSD does not depend on the optical properties of the sample, ELSD detection does not require the sample to have chromophores or fluorescent groups.
The UM-5000 evaporative light scattering detector ensures the formation of a narrow droplet size distribution in the atomization chamber through precise control of air pressure and temperature, greatly reducing the temperature required for droplet evaporation. By adopting a low-temperature evaporation mode, the uniformity of particles is maintained, and it also has good sensitivity to semi volatile substances and thermosensitive compounds.
